Flow-valve Diagnostics for Simple, Point-of-care Analyte Quantitation

We have developed a novel microfluidic system for the simple visual quantitation of target molecules in microliter volumes, for application in point-of-care diagnostics. We constructed microfluidic conduits with a critical dimension (height) of a few micrometers in a deformable material and coated the microchannel surfaces with receptors for the target. When a solution containing the target flowed through a device, the target interacted with receptors on the microchannel surfaces, constricting the channel within the deformable material and stopping flow. The distance the solution traveled before flow ceased was easily observed upon visual inspection and then correlated with the target’s concentration.
